It will change from guitar to guitar, I don't have a line 6 spider, but I guess it has the same as my amp, like 6 different models something like: clean, blues, crunch, metal, insane.

Depending on the output of the guitar start with the blues or crunch or one of those in between, play with the drive until you get a nice distorted sound but still kind of clean, Personally I am not a fan of super high gain, It is sort of like a distorted sound but if I barely hit the string it will sound kind of clean.

Now my guitar for some reason has lots of highs so I bring down the treble and presence, more mids does the trick. Of course add a little reverb and delay to taste.

Try from blues settings and listen your sound and Santana's in parallel, and try to match with the EQ, change pickups, and try to find just right amount of drive. After some time tweaking you should be able to know how to make it similar mate.
