My guitar has both Humbucker and Single coil pickups but i really can't tell the difference in sound between the 2. Am i doing something wrong? Am I Deff?

Turn off your distortion and (perhaps) compression. Clean channel with a little reverb should make it much more obvious a difference. If it doesn't, perhaps a visit to the otolaryngologist for some frequency testing?

Do as jafomatic suggested. You should be able to hear the difference. Make sure you select only single and only humbucker with your pickup selector on the guitar. They should sound different for sure, single coil being more "twangy" "quack" sounding and humbucker being a little bit louder...

ahha, i can see a difference.... i guess it now comes down to a lack of experience in knowing how/when to apply different pickups to music .

Santana talks about one of the differences here:

The thread this was posted in was used to ask about his t-shirt but the content of his lecture in this video is pretty interesting and important. You can use your pickup choice for its tension/resolution as much as you can for other tonal consideration.

Keep in mind that the position where the single coil/humbacker is place makes a difference in tone as well.
