This is a very cool video. It would be very interesting to do something similar for lead playing, I would expect the differences to be bigger.

Very cool idea to do this test. To me the guitar tone didn't change much between players, if one would do a blindfolded test it might be difficult to notice the changes. What was noticeable how different players had different technique and dynamics. Some were playing louder than others and also the way they played the lines (more or less palm muting for example etc).
Having this in mind, if one would want to sound like a specific guitar hero, one just needs similar (same preferably) gear and lots of practice and then it is perfectly achievable. Probably not possible to get it 100% same (for example playing a solo cover), but one can get very close for sure! Terrible test.
With that much gain and the muting ... of course the differences will be minimal at best. Of he didn't get the results he was expecting. He's probably never played a 'clean' toned instrument in his life. *Stuff like this is where the internet displays it's maximum potential for complete suckage. Do it with a Stratocaster set to bridge/middle setting into a deluxe reverb (and maybe a TS set for slight OD) and then lets see where the tone is. I left a comment for Spectre saying I think where differences really make themselves apparent is in lead tones. Bending a note is where the characteristics really come out. He was honest in saying the results weren't as different as he suspected they might be, but realistically, they weren't doing anything but chugging - basically pressing down strings.
