Explaining Harmony Leads

For example, if I were playing a pentatonic minor scale starting at a root note and wanted to play thirds, would I just count three steps up from that root note and start my harmony lead there? Keyboard players have a hard time playing harmonies - while guitarists only have to move the scale shape up/down on the neck, couldn't be easier! For a whacky sound, try minor seconds (one step up) or flat fifths (6 step up).
