Both ways would be cool to add to your practicing routing, yeah.
Eventually you'll start gigging more and more and this need for practicing while standing will become less and less the must, your body will get used to standing position and everything will be much easier to switch from sitting to standing. But until then, go for both ways. -------------------- Youtube

If you feel uncomfortable, I suggest that you practice all the things you have to play live while standing up. This will give your fingers and body a good oportunity to adapt to this position, and will make things easier in the future as well.

A good point of view is what Guthrie Govan says in his book, eg. the right hand shall always have some connection with the guitar, since its unlikely You will stand absolutely still on the stage, and I guess that it also forces You to play more with the wrist than from the elbow, which is actually a good thing.... then of course there may be exceptions where You are needed to lift Your right hand from the guitar.

Its best to practice both ways. I often practice some things standing up and I remember I had a habit of practicing standing for a whole year. Good thing to do when sitting down is to adjust the strap that way to that the guitar stays in almost same position when you do stand up.
